The differentiation of myeloid cells is associated with a marked increase in proteoglycan synthesis. This study investigates the effect of retinoids on proteoglycan synthesis and other properties of the HL60 cell line, a human myelogenous leukemia cell line with both myeloid and monocytoid differentiation potential. When retinoic acid (RA) was added to the culture medium, there was a rapid and reversible inhibition of proteoglycan synthesis, an increase in cell spreading, a decrease in cell adhesiveness and an inhibition of surface phosphatase activity. These findings demonstrate that retinoids can regulate differentiation-associated proteoglycan synthesis in the HL60 cell line. Moreover, these results suggest that the in vivo effects of retinoids on both leukocyte and connective tissue cells may be mediated by a similar mechanism.Q:
